DESCRIPTION
5-Amino-1MQ (5-amino-1-methylquinolinium) is a synthetic small-molecule inhibitor of nicotinamide N-methyltransferase (NNMT), an enzyme that consumes NAD+ precursors and is reported to be elevated in aged adipose tissue, obese skeletal muscle, and certain tumor types. By selectively blocking NNMT, 5-Amino-1MQ is investigated for its ability to preserve cellular NAD+ pools, reduce methylation of nicotinamide to inactive 1-methylnicotinamide (1-MNA), and enhance sirtuin activity and energy metabolism. Originally characterized at the University of Florida by the Stephen O’Rahilly and Robert Kraft groups, the compound has been studied in preclinical research for body composition, adipose tissue lipolysis, glucose homeostasis, skeletal muscle aging, and metabolic disease endpoints.
